Spot test for detection of sickling hemoglobin.

چکیده
We describe a simple, noninstrumental procedure for detecting hemoglobin S in samples of blood on filter paper, with use of small volumes of reagents. The method depends on the differential solubilities of reduced sickling and nonsickling hemoglobin in phosphate solutions (1, 2) or Sickledex reagent’ and the apparent solubilizing effect of urea on reduced hemoglobin S (3). The color of paired phosphate and phosphate-urea hemolysates distinguishes hemoglobin SS from AS and other hemoglobin types.

Rapid solubility test for detection of hemoglobin S.
Several screening methods for detecting sickling hemoglobins are presently used in the clinical laboratory (1-6). The present procedure depends on the insolubility of reduced hemoglobin S in a 2.24 mol/liter phosphate buffer. The commercially available “Sickledex” (Ortho Diagnostics, Raritan, N.J. 08869) is also thought to be based on this principle (6). Our method was compared with the standar...
متن کاملAutomated dithionite test for rapid, inexpensive detection of hemoglobin S and non-S sickling hemoglobinopathies.
Automated adaptations of dithionite and urea-dithionite tube tests are accurate, reliable, inexpensive methods for detecting hemoglobin S. More than 3,000 individuals have been screened (120 determinations per hour; reagent cost, 2 to 4 cents per test). The dithionite reagent consists of potassium phosphate, sodium dithionite, and saponin. When S hemoglobin contacts this reagent, the red cells ...
متن کاملThe Reliability of Sickling and Solubility Tests and Peripheral Blood Film Method for Sickle Cell Disease Screening at District Health Centers in Uganda
Although sickling and solubility tests and peripheral blood film methods are today available for sickle cell disease screening in Uganda, their reliability and ease of applicability have not been determined. This study was therefore carried out to determine the reliability of sickling and solubility tests and peripheral blood film method for screening for SCD in Uganda. This was descriptive lab...
متن کاملEffects of cyanate and 2,3-diphosphoglycerate on sickling. Relationship to oxygenation.
Cyanate and 2,3-diphosphoglycerate (2,3-DPG) both influence the oxygen affinity of hemoglobin. The studies presented here concern the effects of these compounds on the sickling phenomenon. The inhibitory effect of cyanate on sickling is largely due to the fact that it increases the percentage of oxyhemoglobin S at a given oxygen tension. In addition, cyanate inhibits sickling by a mechanism tha...
متن کاملPrevalence of Sickle Cell Trait in the Southern Suburb of Beirut, Lebanon
OBJECTIVE The objective of this study was to assess the prevalence, gender differences, and time trends of Sickle Cell Trait in the Southern Suburb of Beirut, Lebanon, as well as to highlight the importance of screening for Sickle Cell Trait carriers in this population. Another objective was to describe a new screening technique for Sickle Cell Trait carriers. METHODS This was a retrospective...
